Plantaricin W is a two peptide lantibiotic produced by lactobacillus plantarumS34 isolated from bekasam, a traditional fermented food of meat products from Waykanan, Lampung. Plantaricin W is a potential antimicrobial peptide. however, the expression of recombinant plantaricin W has not been reported. To express a recombinant plantaricin WS34, we constructed and expressed the gene encoding plantaricin W in pET-32a by using the T7 RNA polymerase promoter with approximately 33 kDa in size in escherichia coliBL21 (DE3)(pLysS).
Over-expression of proto-oncogene her-2occurs in 20–30% of the total breast cancer cases associated with increased metastatic potential and poor prognosis. It consists of a 620 aa extracellular domain, followed by a 23 aa trans-membrane domain and a 490 aa intracellular domain with tyrosine kinase activity. Treatment using the monoclonal antibody, trastuzumab, was an expensive treatment with an incidence rate of resistance in single use reaches a high percentage of 66-88%. until now, there are continuous efforts for materials or methods improvement for her-2breast cancer treatment. vaccination is an attractive alternative approach to provide protective immunity.
